高通量miRNA表達差異定量檢測陣列 |
MicroRNAs (miRNAs)是一種在轉錄水平調節基因表達的非編碼小RNAs，長度通常在21-23個核苷酸之間。miRNAs在真核生物中高度保守并在細胞通路中起到重要的調節作用。miRNA調節基因的異常表達與癌癥、心血管障礙等多種疾病有關。
miProfile™ miRNA qPCR Arrays主要用于對感興趣組織或細胞中miRNA的表達量進行分析，從而發現與您研究相關的特異miRNAs。每塊96孔板（或384孔板）包含84 條（或360條）已交聯在反應孔中的PCR引物（正向：miRNA特異引物；反向：通用引物），同時包含12個（或24個）用于監控從反轉錄到qPCR反應的每個實驗步驟效率的對照孔。
每條miRNA引物驗證均產生單一擴增產物，融解曲線均呈單一銳鋒，電泳條帶均呈單一條帶，且與目標miRNA相符。由10種人組織混合樣本的總RNA經逆轉錄形成的cDNA作為引物驗證的模板。
使用通用的實時定量反應條件，即可簡便進行高通量的miRNA表達量檢測和分析。逆轉錄試劑All-in-One miRNA First-Strand cDNA Synthesis Kit 和熒光染料SYBR® Green為基礎的定量檢測試劑All-in-One qPCR Mix Kit作為被推薦的RT-qPCR試劑與miProfile miRNA qPCR Array配套使用。該系列試劑均經過優化，具有高的靈敏度、效率和特異性。

miProfile™ Custom miRNA qPCR Arrays 特為定量檢測客戶指定miRNA的表達量差異而設置。每個特定miRNA引物均使用專利算法設計并經過實驗確證。根據檢測miRNA數量和重復次數，GeneCopoeia可提供6種96孔板和8種384孔板的排布方式供客戶選擇。定制產品同樣包含各種不同種類的對照，用以監測樣品質量、RT和PCR反應效率、復孔的重復性等。
